Se sintetizaron y caracterizaron espectroscópicamente derivados aniónicos de metil

Abstract:
Anionic derivatives of methyl β-laminarabioside, methyl β-laminaratrioside and methyl β-maltoside, substituted in the primary hydroxyl groups, were synthesized and characterized spectroscopically. Sulfated laminara-oligosaccharides (DP≥5) were also prepared. The influence of anionic substituents on the glicosidic linkage was studied 6,6’-di-O-sulfated (36 and 38), 6,6’-di-O—phosphated (52 and 53), 6,6’-di-0—succinilated (56 and 57) derivatives of metil β-laminarabioside (13) and metil β-maltoside (17) and the corresponding uronates (60 and 61), as well as unsulfated (5) and sulfated (35) derivatives of laminara-oligosaccharides were chosen as oligosaccharides models. The solution conformation of anionic oligosaccharides has been studied through NMR spectroscopy and molecular mechanics calculations using the MM2 (ɛ: 78, ɛ: 1 D and continuum water model), MM3 (ɛ: 78 D) and AMBER (ɛ: 78 D) force fields. All the experimental evidences from the NOEs based on NOESY and ROESY experiments have been assisted with molecular mechanics calculations. The anionic derivatives, were compared with the parent compounds. Mayor differences were found for fosfated and sulfated derivatives, both in maltosides and laminarabiosides. Qualitative results for laminara-oligosaccharides suggest internal motion mayor restriction for sulfated derivatives.
